Instructions: 1. The female dog must be in a proper state of estrus. The average heat cycle is 18 days and true estrus will usually begin on day 9. This should be verified by a vaginal smear exhibiting a high degree cornified vaginal epithelial cells with absence of neutrophils. Other signs are a receptive female that ‘flags’ her tail or ‘winks’ her vulvar area. Vaginal discharge will change from red to a light amber color. 2. Place the wide end of the latex cone so that it overlaps the rim of the cone holder. Put a small a small amount of J-jelly non-spermicidal lubricating gel inside the latex cone. 3. Remove the plunger rod from the 12cc syringe and put aside for later. Place the small plastic cap tightly on the end of the syringe barrel. Gently put the small end of the latex cone over the top or open end of the syringe barrel. The collection device is now ready. 4. Allow the male to contact the hindquarters of the female and gently massage the sheath until in a full erection is obtained. The bulbourethral gland at the base of the penis should be exposed before a full erection is achieved. The ejaculate consists of three portions. The first portion will be a clear 1 to 3 ml, followed by the sperm rich faction of 1 to 3 ml. The last portion is the prostatic portion of which a large volume may be present. Once you get 2 to 3 ml of the 3rd portion, the collection device is gently removed from the penis. The syringe barrel which has the collected semen in it is removed from the latex cone. The plunger is reattached by gently inserting and tipping the syringe upward. The plastic cap is removed and air is pushed out. Attach the pipette to the syringe containing the semen. A small amount of lubricant should be applied to the pipette. The female’s hindquarters are now slightly elevated (over a chair). The pipette is gently passed first upward toward the tail until over the pubis, the forward to the cervix or where resistance is met. The semen is infused slowly. The tubing is then clamped and the syringe ½ filled with air is infused into the empty pipette of semen. The female should remain with hindquarters elevated for 15 minutes. The AI should be repeated every 2 days until a vaginal smear indicates the end of estrus. The kit can be used again, but needs to be sterilized between each use to avoid disease transmission. |
